Sex-dependent influence of LMAN1 on lung function and allergen-induced airway hyperresponsiveness

چکیده

Abstract We recently reported LMAN1 as a regulatory receptor capable of downregulating NF-kB signaling in response to house dust mite (HDM) vitro. In this follow up work, we have subjected WT and KO mice HDM model allergic airway inflammation assessed lung function hyperreactivity (AHR). Given our previous vitro findings, had expected worsened disease the absence LMAN1. Interestingly, when were stratified by sex, observed that while female, HDM-treated exhibited signs enhanced narrowing closure AHR compared their counterparts, male, showed opposite trend. These sex-dependent effects appeared be localized peripheral airways. additionally characterized type extent immune cell infiltration into highlight those features which might account for gender-biased on setting asthma.

Spred-1 negatively regulates allergen-induced airway eosinophilia and hyperresponsiveness

T helper 2 cytokines, including interleukin (IL)-4, IL-5, and IL-13, play a critical role in allergic asthma. These cytokines transmit signals through the Janus kinase/signal transducer and activator of transcription (STAT) and the Ras-extracellular signal-regulated kinase (ERK) signaling pathways. Although the suppressor of cytokine signaling (SOCS) family proteins have been shown to regulate ...

Detection of allergen-induced airway hyperresponsiveness in isolated mouse lungs.

Airway hyperresponsiveness (AHR) is a hallmark of bronchial asthma. Important features of this exaggerated response to bronchoconstrictive stimuli have mostly been investigated in vivo in intact animals or in vitro in isolated tracheal or bronchial tissues. Both approaches have important advantages but also certain limitations. Therefore, the aim of our study was to develop an ex vivo model of ...

Adrenomedullin insufficiency increases allergen-induced airway hyperresponsiveness in mice.

Adrenomedullin (ADM), a newly identified vasodilating peptide, is reported to be expressed in lungs and have a bronchodilating effect. We hypothesized whether ADM could be involved in the pathogenesis of bronchial asthma. We examined the role of ADM in airway responsiveness using heterozygous ADM-deficient mice (AM+/-) and their littermate control (AM+/+). Here, we show that airway responsivene...

Allergen-induced Bronchial Hyperresponsiveness

Bradykinin (BK) is a peptide mediator generated at sites of inflammation and its effects are mediated through constitutively expressed B 2 receptor or through induction of B 1 receptors. We examined the role of these receptors in bronchial hyperresponsiveness (BHR). Brown-Norway rats sensitized with ovalbumin (OA) and Al(OH) 3 intraperitoneally, were exposed 3 wk later to either saline or OA ae...

Determinants of lung function and airway hyperresponsiveness in asthmatic children.

BACKGROUND Asthma patients exhibit an increased rate of loss of lung function. Determinants to such decline are largely unknown and the modifying effect of steroid therapy is disputed. This cross-sectional study aimed to elucidate factors contributing to such decline and the possible modifying effect of steroid treatment. METHODS We analyzed determinants of lung function and airway hyperrespo...
